Requirements for inspection: To obtain accurate results, the stool must be fresh and the inspection time should not exceed 24 hours. If you check the protozoa trophozoites in the intestines, it is best to check them immediately, or temporarily store them at 35 °C ~ 37 °C. Containers containing feces must be clean, dry, and prevent contamination; feces should not be mixed with urine and other body fluids, so as not to affect the results of the inspection. Inspection process 1. Worm egg inspection: drop a drop of normal saline on a clean glass slide, pick a mung bean-sized fecal block with a cotton swab or toothpick, and spread it evenly in saline; the thickness of the smear is faintly identifiable through the slide. The writing on the screen is appropriate. Generally, it is checked under a low power microscope. If it is observed with a high power microscope, it is necessary to cover the sheet. Attention should be paid to the identification of foreign bodies in eggs and feces. The eggs have a certain shape and size; the surface of the egg shell is smooth and tidy, with an inherent color; the egg contains egg cells or larvae. 2. Protozoal examination (1) Live trophozoite examination: the smear should be thin, and the method should be the same as the worm egg. The closer the temperature is to body temperature, the more obvious the activity of the trophozoites. If necessary, the holding table can be used to maintain the temperature. (2) Encapsulation of iodine staining: The direct smear method is the same as above, and one drop of iodine is used instead of physiological saline. The method of smear application is the same as above. If you need to check the live trophozoites, you can drop a drop of normal saline on the other side of the slide. Apply the stool specimen to the same method as above, and then cover the cover. Check the cyst on the side of the iodine solution in the tablet; check the live trophozoite on the other side. (3) Cryptosporidium oocyst staining examination: The best method at present is Jinan-phenol modified acid-fast staining method. Followed by the gold aminophenol staining method and the improved acid-fast staining method. These three methods can be used for fresh feces or for the preservation of oocysts containing 10% formalin (4 ° C for 1 month).
