Radioallergen Sorption Test (RAST)

The radioallergen adsorbent test (radioallergosorbent test, RAST) combines the purified allergen with the solid phase carrier, adds the serum to be tested and the reference control, and then reacts with the isotopically labeled anti-IgE antibody, and then measures the radioactivity of the solid phase. The content of specific IgE in the serum to be tested was determined by a standard curve, or was positive when the radioactivity of the specimen was higher than the normal number of people plus 3S. Basic Information Specialist classification: skin examination classification: immune examination Applicable gender: whether men and women apply fasting: not fasting Analysis results: Below normal: Normal value: no Above normal: negative: normal. Positive: Abnormal results: The number of nuclide pulses is greater than the normal person's average value (need to subtract the cord blood radioactivity count pulse value) 3.5 times or more as positive. Which item is positive in the test item is sensitive to the allergen and should be avoided as much as possible. Tips: Antihistamine should be discontinued before the test, and adrenal glucocorticoid should be stopped when conditions permit. Normal value Allergens were negative. Clinical significance Abnormal results: It is judged to be positive by the fact that the number of nuclide pulses is larger than the normal person's average value (need to subtract the cord blood radioactivity count pulse value) by 3.5 times or more. Which item is positive in the test item is sensitive to the allergen and should be avoided as much as possible. Need to check the crowd: Asthma, nasal sensitivity, chronic rhinitis, allergic dermatitis and other suspected sensitive constitution. Precautions Taboo before inspection: Antihistamines should be discontinued before the test, and adrenal glucocorticoids should be discontinued when conditions permit. Requirements for inspection: The antigen used should be sterile and of appropriate concentration. The test operation should be accurate; Should pay attention to the presence or absence of systemic reactions, and prepare emergency medicine, just in case. Inspection process Filter paper is used as a carrier. The anti-IgE antibody is coupled to the filter paper activated by cyanogen bromide, and reacted with the serum to be tested and the IgE reference standard; after washing, 125I-labeled anti-human IgE is added, and after washing, the radioactivity of the filter paper sheet is measured. The measured value is positively correlated with the IgE content in the specimen. Not suitable for the crowd There are no taboos. Adverse reactions and risks There are no related complications and hazards.
